New research indicates that females in certain whale species have evolved to have significantly longer lifespans in order to care for their families. The study focussed on five whale species that – along with humans – are the only mammals known to go through menopause. The findings show that females of these whale species that experience menopause live around 40 years longer than other female whales of a similar size. By living longer without extending their “reproductive lifespan” (the years in which they breed), these females have more years to help their children and grandchildren, without increasing the “overlap” period when they compete with their daughters by breeding and raising calves at the same time. This new research shows that – despite being separated by 90 million years of evolution – whales and humans show remarkably similar life histories, which have evolved independently. Reference Permit NMFS-27038 Study Origins and Evolutionary Insights “The process of evolution favors traits and behaviors by which an animal passes its genes to future generations,” said lead author Dr Sam Ellis, from the University of Exeter. “The most obvious way for a female to do this is to breed for the entire lifespan – and this is what happens in almost all animal species. There are more than 5,000 mammal species, and only six are known to go through menopause. So the question is: how and why did menopause evolve? Our study provides some of the answers to this fascinating puzzle.” Menopause is known to exist in five species of toothed whales: short-finned pilot whales, false killer whales, killer whales, narwhals, and beluga whales. As well as outliving females of other similar-sized species, females in these five species outlive the males of their own species. For example, female killer whales can live into their 80s, while males are typically dead by 40. Evolutionary Conditions for Menopause “The evolution of menopause and a long post-reproductive life could only happen in very specific circumstances,” said Professor Darren Croft, of the University of Exeter and Executive Director at the Center for Whale Research “Firstly, a species must have a social structure in which females spend their lives in close contact with their offspring and grand-offspring. Secondly, the females must have an opportunity to help in ways that improve the survival chances of their family. Reference Permit NMFS-27038 Professor Dan Franks, from the University of York, said: “Previous research on menopause evolution has tended to focus on single species, typically humans or killer whales. This study is the first to cross several species, enabled by the recent discovery of menopause in multiple species of toothed whales. Our study provides evidence that menopause evolved by expanding female lifespan beyond their reproductive years, rather than from reduced reproductive lifespan. This is a question that has long been asked in anthropology, but can only be directly answered with a comparative study.” Commenting on parallels with the evolution of menopause in humans, Professor Croft added: “It’s fascinating that we share this life history with a taxonomic group we’re so different from. “Despite these differences, our results show that humans and toothed whales show convergent life history – just like in humans, menopause in toothed whales evolved by selection to increase the total lifespan without also extending their reproductive lifespan.” Reference: “The evolution of menopause in toothed whales” by Samuel Ellis, Daniel W. Credit: Elke Gabriel Human induced pluripotent stem cells (iPSCs) can be used to generate brain organoids containing an eye structure called the optic cup, according to a study published on August 17, 2021, in the journal Cell Stem Cell. The organoids spontaneously developed bilaterally symmetric optic cups from the front of the brain-like region, demonstrating the intrinsic self-patterning ability of iPSCs in a highly complex biological process. “Our work highlights the remarkable ability of brain organoids to generate primitive sensory structures that are light sensitive and harbor cell types similar to those found in the body,” says senior study author Jay Gopalakrishnan of University Hospital Düsseldorf. “These organoids can help to study brain-eye interactions during embryo development, model congenital retinal disorders, and generate patient-specific retinal cell types for personalized drug testing and transplantation therapies.” Many aspects of human brain development and diseases can be studied using 3D brain organoids derived from pluripotent stem cells, which can give rise to all cell types in the body. Researchers previously used human embryonic stem cells to generate the optic cup, which gives rise to the retina—the light-sensitive layer of tissue at the back of the eye. Another study demonstrated that optic-cup-like structures can be generated from iPSCs, which are derived from adult cells that have been genetically reprogrammed back into an embryonic-like pluripotent state. Credit: Gabriel et al./Cell Stem Cell In the past, the production of optic cups from pluripotent stem cells focused on generating the pure retina. Until now, optic cups and other 3D retinal structures had not been functionally integrated into brain organoids. To achieve this feat, Gopalakrishnan and his team modified a protocol they previously developed for turning iPSCs into neural tissue. The human brain organoids formed optic cups, which appeared as early as 30 days and matured as visible structures within 50 days. This time frame parallels that of retinal development in the human embryo and could make certain types of developmental neurobiology experiments more efficient. Across 16 independent batches from four iPSC donors, the researchers generated 314 brain organoids, 72% of which formed optic cups, showing that the method is reproducible. These structures contained diverse retinal cell types, which formed electrically active neuronal networks that responded to light. The optic cup brain organoids also contained lens and corneal tissue and exhibited retinal connectivity to brain regions. “In the mammalian brain, nerve fibers of retinal ganglion cells reach out to connect with their brain targets, an aspect that has never before been shown in an in vitro system,” Gopalakrishnan says. In future studies, they plan to develop strategies to keep the optic cups viable for long time periods, using them to investigate mechanisms that cause retinal disorders. Kobe University’s discovery of new PGC-1⍺ protein variants, which are more active during exercise and can regulate fat burning and energy metabolism, suggests a potential breakthrough in treating obesity through increasing energy expenditure rather than merely reducing caloric intake. New findings highlight PGC-1⍺ variants “b” and “c” as key to improving fat burning and energy metabolism during exercise, offering new avenues for obesity treatment. Some people lose weight slower than others after workouts, and a Kobe University research team found a reason. They studied what happens to mice that cannot produce signal molecules that respond specifically to short-term exercise and regulate the body’s energy metabolism. These mice consume less oxygen during workouts, burn less fat, and are thus also more susceptible to gaining weight. Since the team found this connection also in humans, the newly gained knowledge of this mechanism might provide a pathway for treating obesity. The Link Between Exercise and Fat Burning It is well known that exercise leads to the burning of fat. But for some people, this is much more difficult than for others, casting doubt on whether the mechanism behind losing or gaining weight is as simple as “calories in minus calories out.” Researchers have previously identified a signal molecule, a protein by the name of “PGC-1⍺,” that seems to link exercise and its effects. However, whether an increased amount of this protein actually leads to these effects or not has been inconclusive, since some experiments suggested it while others didn’t. More recently, the Kobe University endocrinologist Wataru Ogawa as well as other researchers found that there are actually a few different versions of this protein. Ogawa explains: “These new PGC-1α versions, called “b” and “c,” have almost the same function as the conventional “a” version, but they are produced in muscles more than tenfold more during exercise, while the a version does not show such an increase.” His team therefore set out to prove the idea that it is the newly discovered versions, and not the previously known one, that regulate the energy metabolism during workouts. The different versions of the signal molecule PGC-1⍺ react to different stimuli. The standard version (“a”) is produced in response to long-term exercise, whereas the alternative versions (“b/c”) are produced in response to short-term exercise or cold exposure. DOI: 10.1016/j.molmet.2024.101968 To do so, the researchers created mice that lack the b and c versions of the signal molecule PGC-1⍺ while they still have the standard a version, and measured the mice’s muscle growth, fat burning, and oxygen consumption during rest and short-term as well as long-term workout. They also recruited human test subjects with and without type 2 diabetes and submitted them to similar tests as the mice, because insulin-intolerant and obese people are known to have reduced levels of the signal molecule. Biological Implications of Protein Variants Ogawa and his team published their results in the journal Molecular Metabolism. They found that, while all versions of the signal molecule cause similar biological reactions, their different levels of production have far-reaching consequences for the organism’s health. The lack of the alternative b and c versions of PGC-1⍺ means that the organism is essentially blind to short-term activity and does not adapt to these stimuli, with the effect that such individuals consume less oxygen and burn less fat during and after workouts. In humans, the research team found that the more the test subjects produced the b and c versions of the signal molecule, the more they consumed oxygen and the less percent body fat they had, across healthy individuals and those with type 2 diabetes. “Thus, the hypothesis that the genes in skeletal muscle determine susceptibility to obesity was correct,” summarizes Ogawa these findings. However, they also found that long-term exercise stimulates the production of the standard a version of PGC-1⍺, and mice that exercised regularly over the course of six weeks exhibited an increase in muscle mass irrespective of whether they could produce the alternative versions of the signal molecule or not. Credit: Wataru Ogawa Long-Term Effects and Cold Tolerance In addition to the production in muscles, the Kobe University team looked at how the production of the different versions of PGC-1⍺ changes in fat tissues, and found no relevant effect in response to exercise. However, since animals also burn fat to maintain body temperature, the researchers also investigated the mice’s ability to tolerate cold. Indeed, they found that the production of the b and c versions of the signal molecule in brown adipose tissue is increased when the animals are exposed to cold, and that the body temperature of individuals that cannot produce these versions dropped significantly under these conditions. On the one hand, this may contribute to these individuals’ having more body fat, but on the other hand, it seems to imply that the b and c versions of the signal molecule may be responsible for metabolic adaptations to short-term stimuli more generally. Potential Treatments for Obesity Ogawa and his team point out that understanding the physiological activity of the different versions of PGC-1⍺ might allow to devise treatment approaches for obesity: “Recently, anti-obesity drugs that suppress appetite have been developed and are increasingly prescribed in many countries around the world. However, there are no drugs that treat obesity by increasing energy expenditure. If a substance that increases the b and c versions can be found, this could lead to the development of drugs that enhance energy expenditure during exercise or even without exercise. Such drugs could potentially treat obesity independently of dietary restrictions.” The team is now conducting research to find out more about the mechanisms that lead to the increased production of the signal molecule’s b and c versions during exercise.
