Credit: NTU Singapore NTU Singapore’s groundbreaking study on ‘mini kidneys’ offers new hope for treating polycystic kidney disease, with minoxidil emerging as a promising therapy. Scientists at Nanyang Technological University, Singapore (NTU Singapore) have successfully grown ‘mini kidneys’ in the lab and grafted them into live mice, revealing new insights into the metabolic defects and a potential therapy for polycystic kidney disease. ‘Mini kidneys,’ or kidney organoids, are kidney-like structures grown in the lab using stem cells. In the study led by NTU’s Lee Kong Chian School of Medicine (LKCMedicine), researchers grew the organoids using skin cells derived from patients with polycystic kidney disease (PKD), a prevalent form of genetic condition that affects 1 in 1000 individuals across all ethnicities.[1] People with PKD often progress to end-stage kidney disease between their 50s and 60s, with the standard treatment options available being dialysis or a kidney transplant. However, dialysis significantly compromises a patient’s quality of life, while a transplanted kidney can be challenging to acquire. One other option is the Food and Drug Administration (FDA) approved drug Tolvaptan, which is very costly and has severe side effects on the liver. Credit: NTU Singapore To address the need for more effective treatment for PKD patients, the NTU research team sought to better understand the disease by engrafting their newly developed mini kidneys into mice. Previous studies were conducted on mini kidneys grown in a dish, which could only partly mimic the kidney structure and function. The NTU scientists engrafted the mini kidneys into live mice to comprehensively replicate the pathological features of kidney disease, including blood flow, fluid movement (tubular fluid) and cellular communication with other organs. Lead investigator Assistant Professor Xia Yun at LKCMedicine said, “Engrafting the kidney organoid in mice provided us with a physiologically sophisticated approach to studying polycystic kidney disease as we were able to successfully emulate critical disease characteristics similar to those observed in human kidney patients.” Critical disease characteristics included abnormalities like the spontaneous formation of cysts in the kidneys and the subsequent damage to its tiny tubes. Credit: NTU Singapore Innovations in PKD Treatment Research In their study, reported in the scientific journal Cell Stem Cell, the NTU research team said that they believed their engrafted mini kidneys were high quality because cysts sustained without extra stress stimulation or chemicals, even after they were removed from the live mice for further investigations in a dish. In contrast, previous kidney organoids grown in a dish cannot form cysts without stress stimulation. Co-investigator Assistant Professor Foo Jia Nee at LKCMedicine said, “The similarity between the disease manifestation observed in our engrafted mini kidney model and the real-life experiences of polycystic kidney disease patients suggest that growing kidney organoids and engrafting them into live mice could be beneficial in studying the disease and a useful tool to test new treatments.” Metabolic Defects in Polycystic Kidney Disease Scientists have long known that abnormalities in a structure on kidney cells, or the primary cilium, cause cysts to form in kidneys. However, tests to understand the regulatory mechanism and relationship between the primary cilium and cell metabolism (autophagy) in live mice with PKD, have not been possible until now. By studying the development of PKD in live mice and testing cellular pathways, researchers found evidence that boosting autophagy could reduce the severity of cysts in the mini kidney. After establishing that boosting autophagy could reduce cysts, the NTU scientists shortlisted 22 drugs known for their effects on cell metabolism and tested them in the lab. Results showed that minoxidil, a clinical drug widely used to cure hypertension and hair loss, effectively reduced cyst formation in the novel mouse model. Future Implications and Studies Asst Prof Xia Yun said, “Our study has demonstrated how cysts in polycystic diseased kidneys can be reduced by boosting autophagy, suggesting that this could be a promising treatment for PKD. Moreover, the proven clinical safety of minoxidil may allow it to be quickly re-purposed to treat PKD patients in clinic. However, more research will be needed to establish this potential.” Commenting as an independent expert, Associate Professor Ng Kar Hui, Senior Consultant, Division of Paediatric Nephrology, Dialysis and Renal Transplantation, Department of Paediatrics, Khoo Teck Puat – National University Children’s Medical Institute, National University Hospital, said, “Polycystic kidney disease is one of the biggest causes of chronic kidney diseases among adults. An effective treatment may potentially ameliorate the rising numbers of people with kidney failure in Singapore. The establishment of such models in live organisms brings us one step closer to finding more treatment options. In future studies, the NTU team will test the efficacy of minoxidil and adapt the mini kidney models to investigate other burgeoning kidney diseases without a strong genetic underpinning, such as diabetic kidney disease. The giant water bug, a large and potentially painful predator, has been spotted for the first time in Cyprus, sparking interest and investigation by scientists. This finding underscores Cyprus’s rich biodiversity and highlights the importance of citizen science in monitoring environmental changes and species migrations. Credit: Hadjiconstantis et al. Cyprus, known for its stunning, sun-kissed shores, stands as a Mediterranean biodiversity haven. Yet, it’s the island’s picturesque beaches, rather than its rich insect life, that usually capture attention. However, the keen eyes and curiosity of some beachgoers have provided important information about a species never recorded before on the island: the giant water bug, also known as a toe biter. Scientists Michael Hadjiconstantis from the Association for the Protection of Natural Heritage and Biodiversity of Cyprus, Iakovos Tziortzis from the Ministry of Agriculture, Rural Development and Environment of Cyprus, and Kadir Bogaç Kunt from the Cyprus Wildlife Research Institute collected information and specimens from an increasing number of records of the giant water bug on the east coastline of the island in late spring and summer of 2020 and 2021. The species, known for inhabiting ponds and slowly moving freshwaters, had never been recorded on the island before, although established populations are known in adjacent Mediterranean countries such as Greece, Turkey, and Israel. The Giant Water Bug: A Formidable Predator Also referred to as the Electric light bug, this giant water bug is described as a vicious hunter, praying on invertebrates, fish, turtles, and even birds. What is even scarier is that, as the largest European true bug and the largest European water insect, measuring up to 12 cm, it has a reputation for inflicting very painful bites when handled carelessly. Its appearance, mainly on the eastern coastal front of the island, was initially recorded by swimmers, who were surprised by the fearsome looks and size of the bug. They either directly contacted the experts or published photos and videos online, mainly on Facebook groups related to biodiversity. The authors collected some of the specimens for further study. They also proceeded with an extensive online search on relevant online observation platforms (i.e. iNaturalist) in order to track any other reports of the species on the island. In addition, they sampled nearby wetlands, but did not spot the bug. End to end, a total of seven sightings were eventually recorded: five from social media and two after direct communication with the author team. Two specimens were obtained and examined morphologically to verify the species. The observations were recorded in a research article in the open-access journal Travaux du Muséum National d’Histoire Naturelle “Grigore Antipa”. Having in mind that the toe biter is an iconic species accompanied by creepy stories concerning its encounters with humans, the authors assume that it is unlikely that it had gone unnoticed for too long. They consider it possible that several migration events might have been triggered in a short period of time by nearby countries hosting the species, such as Israel, Lebanon, and Syria. The specimens could have been transferred by wind or sea currents, as assumed by other researchers, or could have been driven by a decrease in food resources in their initial area of distribution. Despite the number of recordings in a short timeframe, no conclusions can be drawn for the moment on the establishment of a population of the species on the island. This is further to be investigated, and as the initial recording of the species, citizen science can have an important role in this. A tiny clam, previously only known from fossil records, has been discovered alive in the tidepools of Santa Barbara by researchers from the University of California, Santa Barbara and the Santa Barbara Museum of Natural History. The discovery of a new species is always exciting, but it can be even more so when a species thought to be extinct is found alive. Researchers from the University of California, Santa Barbara and the Santa Barbara Museum of Natural History have made just such a discovery at Naples Point, where they found a small clam that had previously only been known from fossil records. The researchers’ findings have recently been published in the journal Zookeys. “It’s not all that common to find alive a species first known from the fossil record, especially in a region as well-studied as Southern California,” said co-author Jeff Goddard, a research associate at UC Santa Barbara’s Marine Science Institute. “Ours doesn’t go back anywhere near as far as the famous Coelacanth or the deep-water mollusk Neopilina galatheae — representing an entire class of animals thought to have disappeared 400 million years ago — but it does go back to the time of all those wondrous animals captured by the La Brea Tar Pits.” A Tiny Clam with a Surprising History In November 2018, during a low tide at Naples Point, Goddard was looking for nudibranch sea slugs when he noticed a pair of small, translucent bivalves. The shells of these bivalves were only 10 millimeters long, but when they extended and began waving a bright white-striped foot that was longer than their shell, Goddard realized that he had never seen this species before. Despite his many years of experience studying intertidal habitats in California, including a significant amount of time at Naples Point, this discovery surprised Goddard. Credit: Jeff Goddard With quality images in hand, Goddard decided not to collect the animals, which appeared to be rare. After pinning down their taxonomic family, he sent the images to Paul Valentich-Scott, curator emeritus of malacology at the Santa Barbara Museum of Natural History. “I was surprised and intrigued,” Valentich-Scott recalled. “I know this family of bivalves (Galeommatidae) very well along the coast of the Americas. This was something I’d never seen before.” He mentioned a few possibilities to Goddard, but said he’d need to see the animal in-person to make a proper assessment. So, Goddard returned to Naples Point to claim his clam. But after two hours combing just a few square meters, he still hadn’t caught sight of his prize. The species would continue to elude him many more times. Nine trips later, in March 2019, and nearly ready to give up for good, Goddard turned over yet another rock and saw the needle in the haystack: A single specimen, next to a couple of small white nudibranchs and a large chiton. Valentich-Scott would get his specimen at last, and the pair could finally set to work on identification. Credit: Jeff Goddard Linking the Discovery to a Fossil Species Valentich-Scott was even more surprised once he got his hands on the shell. He knew it belonged to a genus with one member in the Santa Barbara region, but this shell didn’t match any of them. It raised the exciting possibility that they had found a new species. “This really started ‘the hunt’ for me,” Valentich-Scott said. “When I suspect something is a new species, I need to track back through all of the scientific literature from 1758 to the present. It can be a daunting task, but with experience it can go pretty quickly.” The two researchers decided to check out an intriguing reference to a fossil species. They tracked down illustrations of the bivalve Bornia cooki from the paper describing the species in 1937. It appeared to match the modern specimen. If confirmed, this would mean that Goddard had found not a new species, but a sort of living fossil. It is worth noting that the scientist who described the species, George Willett, estimated he had excavated and examined perhaps 1 million fossil specimens from the same location, the Baldwin Hills in Los Angeles. That said, he never found B. cooki himself. Rather, he named it after Edna Cook, a Baldwin Hills collector who had found the only two specimens known. Valentich-Scott requested Willett’s original specimen (now classified as Cymatioa cooki) from the Natural History Museum of Los Angeles County. This object, called the “type specimen,” serves to define the species, so it’s the ultimate arbiter of the clam’s identification. Meanwhile, Goddard found another specimen at Naples Point — a single empty shell in the sand underneath a boulder. After carefully comparing the specimens from Naples Point with Willett’s fossil, Valentich-Scott concluded they were the same species. “It was pretty remarkable,” he recalled. Small size and cryptic habitat notwithstanding, all of this begs the question of how the clam eluded detection for so long. “There is such a long history of shell-collecting and malacology in Southern California — including folks interested in the harder to find micro-mollusks — that it’s hard to believe no one found even the shells of our little cutie,” Goddard said. Environmental Factors and Distribution He suspects the clams may have arrived here on currents as planktonic larvae, carried up from the south during marine heatwaves from 2014 through 2016. These enabled many marine species to extend their distributions northward, including several documented specifically at Naples Point. Depending on the animal’s growth rate and longevity, this could explain why no one had noticed C. cooki at the site prior to 2018, including Goddard, who has worked on nudibranchs at Naples Point since 2002. “The Pacific coast of Baja California has broad intertidal boulder fields that stretch literally for miles,” Goddard said, “and I suspect that down there Cymatioa cooki is probably living in close association with animals burrowing beneath those boulders.
