Weill Cornell Medicine scientists have discovered that astrocyte receptors can have opposite effects on cognitive function in male and female preclinical models. Published in Cell Reports, the study reveals that astrocytes play a crucial role in sex-specific brain mechanisms. Focusing on the mGluR3 receptor, the research demonstrated that increasing its levels enhanced memory in older females while reducing it impaired memory in young females. Conversely, in males, reducing mGluR3 enhanced memory. These findings suggest that therapeutics targeting astrocytic receptors may need to be evaluated for sex-specific effects, highlighting the importance of considering biological sex in neurological research. Research from Weill Cornell Medicine reveals that astrocyte receptors impact cognitive functions differently in males and females, suggesting a need for sex-specific approaches in developing treatments targeting these brain cells. Scientists at Weill Cornell Medicine have discovered the first evidence that receptors in astrocytes, brain cells that support and regulate neurons, can have contrasting effects on cognitive function in male and female preclinical models. This research highlights the role of astrocytes in contributing to gender-specific brain mechanisms. While many studies have tested the behavioral effects of astrocytic receptors, none of them have addressed whether biological sex plays a role and most have tested only males. This study, published on May 24 in Cell Reports, challenges the long-standing assumption that astrocytic signaling has similar cognitive effects in both sexes. “Our study reveals that previously reported cognitive effects in males can’t be extrapolated to females,” said Dr. Anna G. Orr, the Nan and Stephen Swid Assistant Professor of Frontotemporal Dementia Research and an assistant professor of neuroscience in the Feil Family Brain and Mind Research Institute and the Helen and Robert Appel Alzheimer’s Research Institute at Weill Cornell Medicine. Changes in astrocytic receptors are seen in various neurological conditions with known sex differences, including neurodegenerative disorders, schizophrenia, stroke, and epilepsy. However, the mechanisms promoting sex differences remain poorly understood. How do Male and Female Brains Differ? In the study, Dr. Samantha M. Meadows, first author and former graduate student in the Orr lab, focused on mGluR3, a predominant glutamate receptor in astrocytes and a top-altered gene in dementia. The team used gene editing and stimulation of engineered receptors in animal models to selectively manipulate astrocytes and examine the effects of mGluR3 and related receptors on learning, memory, and other cognitive and behavioral outcomes. The researchers found that increasing astrocytic mGluR3 levels enhanced memory in older females and reducing these levels was sufficient to impair memory in young females, demonstrating that mGluR3 promotes memory recall in females. However, in males, reducing mGluR3 enhanced memory, and increasing the receptor had no effects. “Interestingly, the cognitive impact of these receptors is not conserved among sexes,” Dr. Meadows said. Credit: Orr Lab To understand if these divergent effects were unique to mGluR3 or reflected a broader feature of astrocytic receptor signaling, Dr. Meadows worked with co-author Dr. Adam L. Orr, an assistant professor of research in neuroscience in the Brain and Mind Research Institute and the Appel Alzheimer’s Disease Research Institute, to selectively stimulate different astrocytic receptors while mice performed tasks involving learning and memory. To their surprise, the team found further evidence that receptor activation caused either memory enhancement or impairment, depending on biological sex. “Normal brain function seems to require a sex-specific balance in astrocytic signaling,” Dr. Adam Orr said. This study suggests that mGluR3 modulators being developed for treating disorders such as schizophrenia and anxiety may need further study to assess their impact on different sexes. “Therapeutics influencing astrocytic receptors may cause sex-specific cognitive effects in part due to the divergent roles of astrocytes in males and females,” said Dr. Anna Orr. The lab is investigating what may cause the differential effects and if other brain functions are also changed in a sex-specific way. A new study reveals that SMC proteins not only loop DNA but also twist it, a process conserved across different organisms. This finding is key to understanding DNA organization and its impact on gene expression and chromosome integrity, with implications for genetic disorders. Scientists from the Kavli Institute at Delft University of Technology and the IMP Vienna BioCenter have uncovered a new property of the molecular motors that organize our chromosomes. Six years ago, they discovered that SMC motor proteins create long loops in DNA. Now, they’ve found that these proteins also introduce significant twists into the loops they form. This discovery deepens our understanding of chromosome structure and function, shedding light on how DNA is managed within cells. It also offers valuable insight into how disruptions in DNA twisting and looping can contribute to health issues, including developmental disorders like cohesinopathies. The researchers published their findings in Science Advances. DNA Compaction and Cellular Function Imagine trying to fit two meters of rope into a space smaller than the tip of a needle. That’s the task every cell in your body faces when packing its DNA into its microscopic nucleus. To accomplish this, nature has developed remarkable strategies. DNA is twisted into tight coils upon coils, forming structures called supercoils (see pictures for a visualization), and wrapped around specialized proteins for efficient storage. When genetic information is needed, specific sections of DNA are temporarily unpacked and read. During cell division, the DNA must fully unwind, duplicate, and separate into two new cells. This process is controlled by specialized protein machines known as SMC complexes (Structural Maintenance of Chromosomes). These molecular motors create long loops in the DNA, which play a critical role in regulating chromosome structure and function. Researchers at Delft and other institutions recently discovered that these DNA loops are central to how chromosomes organize and operate—an essential breakthrough in understanding cellular biology. Innovations in DNA Research In the lab of Cees Dekker at TU Delft, postdocs Richard Janissen and Roman Bath now provide clues that help to crack this puzzle. They developed a new way to use ‘magnetic tweezers’ by which they could watch individual SMC proteins make looping steps in DNA. Importantly, they were also able to resolve if the SMC protein would change the twist in the DNA. And strikingly, the team found that it did: the human SMC protein cohesin does indeed not only pull DNA into a loop, but also twists the DNA in a left-handed way by 0.6 turns in each step of creating the loop. Similar SMC proteins in yeast behave the same way. Strikingly, all the various types of SMC proteins from human and yeast add the same amount of twist – they turn DNA 0.6 times at every at every DNA loop extrusion step. This shows that the DNA extrusion and twisting mechanisms stayed the same for very long times during evolution. No matter whether DNA is looped in humans, yeast, or any other cell – nature employs the same strategy. Implications for Genetic Research and Health These new findings will provide essential clues for resolving the molecular mechanism of this new type of motor. Additionally, they make clear that DNA looping also affects the supercoiling state of our chromosomes, which directly affects processes like gene expression. Finally, these SMC proteins are related to various diseases such as Cornelia de Lange Syndrome, and a better understanding of these processes is vital for tracking down the molecular origins of these serious illnesses. Researchers are now able to wirelessly record the directly measured brain activity of patients living with Parkinson’s disease and to then use that information to adjust the stimulation delivered by an implanted device. Direct recording of deep and surface brain activity offers a unique look into the underlying causes of many brain disorders; however, technological challenges up to this point have limited direct human brain recordings to relatively short periods of time in controlled clinical settings. This project, published in the journal Nature Biotechnology, was funded by the National Institutes of Health’s Brain Research Through Advancing Innovative Neurotechnologies (BRAIN) Initiative. “This is really the first example of wirelessly recording deep and surface human brain activity for an extended period of time in the participants’ home environment,” said Kari Ashmont, Ph.D., project manager for the NIH BRAIN Initiative. “It is also the first demonstration of adaptive deep brain stimulation at home.” Deep brain stimulation (DBS) devices are approved by the U. S. Food and Drug Administration for the management of Parkinson’s disease symptoms by implanting a thin wire, or electrode, that sends electrical signals into the brain. In 2018, the laboratory of Philip Starr, M.D., Ph.D. at the University of California, San Francisco, developed an adaptive version of DBS that adapts its stimulation only when needed based on recorded brain activity. In this study, Dr. Starr and his colleagues made several additional improvements to the implanted technology. “This is the first device that allows for continuous and direct wireless recording of the entire brain signal over many hours,” said Dr. Starr. “That means we are able to perform whole brain recording over a long period of time while people are going about their daily lives.” The implications of this type of recording are significant. The brain activity patterns (neural signatures) normally used to identify problems such as Parkinson’s disease symptoms have traditionally been recorded in clinical settings over short periods of time. This new technology makes it possible to validate those signatures during ordinary daily activities. “If you ever hope to use in-hospital recordings to modify a disease state through adaptive stimulation, you must show that they are also valid in the real world,” said Dr. Starr. Another advantage to recording over long periods of time is that distinct changes in brain activity (biomarkers) that could predict movement disorders can now be identified for individual patients. Ro’ee Gilron, Ph.D., a postdoctoral scholar in Dr. Starr’s lab and first author of this study, explained that this allows for a level of customized DBS treatment that was impossible to achieve previously. “Because we are able to build a biomarker library for each patient, we can now program each DBS unit according to a patient’s individual needs,” said Dr. Gilron. “This includes personalized stimulation programs that adapt as the patient’s needs change throughout the day.” One important consideration that arises is the ethical implication of (nearly) all-day brain recording. Since its beginning, the NIH BRAIN Initiative has recognized the importance of addressing potential ethical considerations pertaining to the development and use of devices that record or modulate brain activity. For instance, the NIH BRAIN Neuroethics Working Group is a group of experts in neuroethics and neuroscience that serves to provide the NIH BRAIN Initiative with input relating to neuroethics — a field that studies the ethical, legal, and societal implications of neuroscience. Alongside funding for neurotechnology research, the Initiative also funds research on the ethical implications of advancements in neurotechnology. “We have had patients approach us with concerns regarding privacy,” said Dr. Starr. “Although we are not at the point where we can distinguish specific normal behaviors from brain activity recording, it is an absolutely legitimate concern. We have told patients to feel free to remove their wearable devices and to turn off their brain recordings whenever they engage in activities they would like to keep private.” The patients were also invited to participate in NIH BRAIN Initiative-funded neuroethics projects looking to identify concerns about this new technology (MH114860). In addition, individuals who opted out of the implant project were interviewed about their decision. As recommended by a recent BRAIN 2.0 neuroethics report, this information will be used to develop ethical guidelines and protocols for future projects to achieve a healthy balance between discovery and privacy. One unforeseen benefit of this study was that, because it required little to no direct contact with clinicians following surgery, it was ideally suited for the social distancing that is crucial during the COVID-19 pandemic. The technologies used for remote patient monitoring and telehealth were originally designed for the convenience of study subjects, but they have broader applications to other research projects that have been stalled due to COVID-19. “The technologies we developed and used to communicate and work remotely with our patients can also allow those who do not live close to a clinic to receive ‘over the air’ updates for their devices and telehealth visits from their neurologists as they manage increasingly complex DBS devices,” said Dr. Gilron. The importance of studying behavior in a natural environment such as the home as it relates to neural activity was emphasized in a recent BRAIN 2.0 neuroscience report. Dr. Ashmont stressed that this study is a significant step in that direction and is going to help scientists understand not only disorders but also the neural representation of behaviors in general.
