Argentine Ant (Linepithema humile). Detecting environmental DNA of invasive ant species in soil samples. Researchers have successfully detected the environmental DNA (eDNA)[1] of the Argentine ant[2] in surface soil samples from sites on Kobe’s Port Island and in Kyoto’s Fushimi District, two areas that have a long history of destruction caused by this invasive species. The research group included then graduate student YASASHIMOTO Tetsu and Associate Professor MINAMOTO Toshifumi of Kobe University’s Graduate School of Human Development and Environment, Visiting Professor OZAKI Mamiko of the Graduate School of Engineering, and NAKAJIMA Satoko, formally of the Kyoto Prefectural Institute of Public Health and Environment. This method can be used to enable scientists to easily gain an accurate understanding of the habitat distribution and hotspots for globally invasive ant species[3], such as the fire ant, which cause significant damage. Combining this method with pest control plans against invasive ant species will contribute towards the formulation of targeted measures and successful elimination results. These research results are due to be published in Scientific Reports today (May 26, 2021). Main points Invasive ant species are causing serious damage worldwide. Early detection and rapid elimination is essential for controlling their populations. However, current pest control methods involve a series of direct detection techniques (such as visual observation, capture, classification, elimination, follow-up observation and evaluation), which require specialist knowledge, labor and time. This is inefficient considering the widespread damage caused worldwide by these invasive species. By focusing on one species that is difficult to eradicate (the Argentine ant), the group demonstrated that eDNA analysis can provide a useful tool for observing and evaluating the invasion, establishment and proliferation of invasive ant species. The research group developed an Argentine ant-specific real-time PCR assay. Using this new assay, they successfully and highly accurately detected eDNA originating from this species in surface samples collected from invasion sites. The researchers compared the presence of eDNA with the last decade of pest control records. They demonstrated the efficiency of eDNA analysis for monitoring populations of the target species and reported for the first time that this method could lead to rapid improvements in the accuracy and effectiveness of invasive ant extermination. Research Background In the midst of globalization, the transport of goods and commodities between nations is increasing. Consequently, the arrival, subsequent establishment and widespread proliferation of invasive ant species that are inadvertently transported to other countries has developed into a worldwide problem. In Japan, Argentine ant (Figure 1A) colonies continue to proliferate widely, and the invasion and establishment of highly poisonous fire ant populations have also been reported. The research group chose two areas that have a long history of damage caused by the Argentine ant; 1. Kyoto’s Fushimi district and 2. Kobe’s Port Island. 1. Fushimi has continuously used insecticide measures for almost 10 years, and has had consistent success in suppressing Argentine ant populations in built-up areas. 2. Although the Argentine ant first invaded Kobe over 20 years ago, the situation has yet to change for the better. In cases like 1, where insecticide-based methods have been carried out for a long period of time, it is difficult to stop the usage of insecticide on a non-scientific basis, even though this presents a problem from the perspective of conserving the natural environment and ecosystem. In cases like 2, there are no pest control plans in place because the extent of the species’ distribution is not readily understood. Environmental DNA analysis is a biological monitoring method that was introduced into the fields of ecology and conservation biology around 2008. It has brought about revolutionary changes in the conduction of biological surveys, especially in aquatic environments. It is hoped that applying this technique to surface soil samples instead of water samples will play a key role in eliminating invasive ants, providing a breakthrough solution to this problem. Research Methodology and Findings Argentine ant-specific DNA assay The researchers designed a real-time PCR assay specific to the Argentine ant and experimentally confirmed the specificity of the assay. The researchers then performed the following experiments using this method. Selection of surface sample sites and their respective pest control histories Surface samples were collected from a total of four sites (FM-1 to 4) in the Fushimi district of Kyoto in order to test them for the eDNA of the Argentine ant, as shown in Figure 1B. The characteristics of efforts to control invasive ant species over the past decade are different for each site (Figure 2). Detecting the eDNA of Argentine ants using surface soil samples Environmental DNA from the Argentine ant was found in the samples from the FM-1 and FM-2 sites, where their presence had previously been confirmed by traditional bait trap surveys. However, eDNA from this invasive species was not detected in the samples taken from sites FM-3 and FM-4. These results did not contradict those obtained using the bait trap method, suggesting that eDNA analysis can be considered more accurate than surveys based on visual observations (Table 1). In addition, native ant species were observed at each of the survey sites. At sites FM-1 and 2, where Argentine ant eDNA was detected, 7 and 3 native species were observed, whereas at sites FM-3 and 4, where the invasive ant’s DNA was not detected, 15 and 10 native species were found respectively. By comparing and analyzing the results of environmental DNA analysis of various ant species with the habitat data of Argentine ant and native ant species accumulated over the years, a method for estimating ant habitat by environmental DNA can be developed for practical use. In this way, eDNA could provide a scientific basis from which to reconsider previous ineffective pest control methods and to eradicate issues relating to the continuance of pest control and confirming extermination. 2. The environmental DNA method is a fundamental, all-purpose technique, which enables further research into other invasive ant species, such as fire ants, to be conducted in the same way. 3. A policy model for the control of invasive species, based on the Sustainable Development Goal (SDG) to ‘Conserve and restore terrestrial ecosystems and halt biodiversity loss’, can be drawn up and implemented, beginning with invasive ant species (including Argentine ant and fire ant). Glossary Environmental DNA (eDNA): This is DNA originating from organisms that is found in the environment, such as in the water or on the ground. Environmental DNA analysis allows researchers to comprehensively identify species that reside in an area, even if they no longer live there. This in turn enables them to estimate and gain an understanding of the target species’ distribution. Environmental DNA surveys are used in a wide range of research fields, including species conservation, ecology, taxonomy, microbiology and palaeontology. Argentine ant (Linepithema humile): The Argentine ant is part of the Dolichoderinae subfamily and belongs to the Hymenoptera order. It originated in South America. The IUCN (International Union for Conservation of Nature and Natural Resources) ranks it as one of the world’s worst 100 invasive species. It is also designated as one of Japan’s worst 100 invasive species. It was first found in Japan in 1993 in Hatsukaichi City (Hiroshima Prefecture) and has since been reported in 11 prefectures. It is a very difficult species to eradicate once it has invaded an area, as the ant establish supercolonies with multiple queens. Invasive ant species: These species have spread beyond their country of origin and have established themselves in countries worldwide. They invade native species’ habitats, as well as the dwellings of people and livestock; posing a threat to living environments, in addition to health and safety. These invasive ant species drive out and eradicate the former occupants of an area, which leads to a loss of biodiversity and ecosystem destruction. MIT scientists found that the protein perlecan, found in both flies and humans, is vital for maintaining the structural integrity of neuronal axons. Without it, axons can break, leading to the death of synapses. Scientists find a protein common to flies and people is essential for supporting the structure of axons that neurons project to make circuit connections. In a study conducted by MIT’s Picower Institute for Learning and Memory, researchers found that a protein named perlecan plays a crucial role in maintaining the structural integrity of neurons. Perlecan is part of the extracellular matrix that surrounds cells and helps them develop in a supportive, yet non-rigid environment. The study revealed that, without perlecan, the axons (long projections of neurons used for connection) can break apart during development, leading to the death of synapses (neuronal connections). Perhaps the most obvious feature of a neuron is the long branch called an axon that ventures far from the cell body to connect with other neurons or muscles. If that long, thin projection ever seems like it could be vulnerable, a new MIT study shows that its structural integrity may indeed require the support of a surrounding protein called perlecan. Without that protein in Drosophila fruit flies, researchers at The Picower Institute for Learning and Memory found axonal segments can break apart during development and the connections, or synapses, that they form end up dying away. Perlecan helps make the extracellular matrix, the proteins, and other molecules that surround cells, stable and flexible so that cells can develop and function in an environment that is supportive without being rigid. “What we found was that the extracellular matrix around nerves was being altered and essentially causing the nerves to break completely. Broken nerves eventually led to the synapses retracting,” says study senior author Troy Littleton, the Menicon Professor in MIT’s departments of Biology and Brain and Cognitive Sciences. MIT researchers have found that a protein called Perlecan is key for sustaining the structural integrity of neuronal axons. Mutations that reduce, but don’t eliminate, perlecan can cause Schwartz-Jampel syndrome, in which patients experience neuromuscular problems and skeletal abnormalities. The new study may help explain how neurons are affected in the condition, Littleton says, and also deepen scientists’ understanding of how the extracellular matrix supports axon and neural circuit development. Ellen Guss PhD ’23, who recently defended her doctoral thesis on the work, led the research published on June 8 in the journal eLife. At first she and Littleton didn’t expect the study to yield a new discovery about the durability of developing axons. Instead, they were investigating a hypothesis that perlecan might help organize some of the protein components in synapses that fly nerves develop to connect with muscles. But when they knocked out the gene called “trol” that encodes perlecan in flies, they saw that the neurons appeared to “retract” many synapses at a late stage of larval development. Proteins on the muscle side of the synaptic connection remained, but the neuron side of the connection withered away. That suggested that perlecan had a bigger role than they first thought. The Neural Lamella and Its Protective Function Indeed, the authors found that the perlecan wasn’t particularly enriched around synapses. Where it was pronounced was in a structure called the neural lamella, which surrounds axon bundles and acts a bit like the rubbery cladding around a TV cable to keep the structure intact. That suggested that a lack of perlecan might not be a problem at the synapse, but instead causes trouble along axons due to its absence in the extracellular matrix surrounding nerve bundles. Littleton’s lab had developed a technique for daily imaging of fly neural development called serial intravital imaging. They applied it to watch what happened to the fly axons and synapses over a four-day span. They observed that while fly axons and synapses developed normally at first, not only synapses but also whole segments of axons faded away. They also saw that the farther an axon segment was from the fly’s brain, the more likely it was to break apart, suggesting that the axon segments became more vulnerable the further out they extended. Looking segment by segment, they found that where axons were breaking down, synapse loss would soon follow, suggesting that axon breakage was the cause of the synapse retraction. “The breakages were happening in a segment-wide manner,” Littleton says. “In some segments the nerves would break and in some they wouldn’t. Whenever there was a breakage event, you would see all the neuromuscular junctions (synapses) across all the muscles in that segment retract.” When they compared the structure of the lamella in mutant versus healthy flies, they found that the lamella was thinner and defective in the mutants. Moreover, where the lamella was weakened, axons were prone to break and the microtubule structures that run the length of the axon would become misdirected, protruding outward and becoming tangled up in dramatic bundles at sites of severed axons. In one other key finding, the team showed that perlecan’s critical role depended on its secretion from many cells, not just neurons. Blocking the protein in just one cell type or another did not cause the problems that total knockdown did, and enhancing secretion from just neurons was not enough to overcome its deficiency from other sources. Rigidity vs. Flexibility Altogether, the evidence pointed to a scenario where lack of perlecan secretion caused the neural lamella to be thin and defective, with the extracellular matrix becoming too rigid. The further from the brain nerve bundles extended, the more likely movement stresses would cause the axons to break where the lamella had broken down. The microtubule structure within the axons then became disorganized. That ultimately led to synapses downstream of those breakages dying away because the disruption of the microtubules means the cells could no longer support the synapses. “When you don’t have that flexibility, although the extracellular matrix is still there, it becomes very rigid and tight and that basically leads to this breakage as the animal moves and pulls on those nerves over time,” Littleton says. “It argues that the extracellular matrix is functional early on and can support development, but doesn’t have the right properties to sustain some key functions over time as the animal begins to move and navigate around. Women experience temporary synchronization of their menstrual cycles with the luminance and gravimetric cycles of the Moon. An analysis of long-term menstrual cycle records kept by 22 women for up to 32 years shows that women with cycles lasting longer than 27 days intermittently synchronized with cycles that affect the intensity of moonlight and the moon’s gravitational pull. This synchrony was lost as women aged and when they were exposed to artificial light at night. The researchers hypothesized that human reproductive behavior may have been synchronous with the moon during ancient times, but that this changed as modern lifestyles emerged and humans increasingly gained exposure to artificial light at night. While previous research suggests that women with menstrual cycles that most closely match lunar cycles may have the highest likelihood of becoming pregnant, lunar influence on human reproduction remains a controversial subject. To address this long-standing mystery, Charlotte Helfrich-Förster and colleagues examined long-term data on the onset of menstrual cycles spanning an average of 15 years, including records from 15 women age 35 or younger and 17 women over 35. To uncover any times during which the women’s menstrual cycles occurred in sync with lunar cycles, the researchers displayed the data as raster plots (graphs that show time-based relations) along with fluctuations in the moon’s cycles. The researchers found that most women’s menstrual cycles aligned with the synodic month (the time it takes for the moon to cycle through all its phases) at certain intervals. Menstrual cycles also aligned with the tropical month (the 27.32 days it takes the moon to pass twice through the same equinox point) 13.1% of the time in women 35 years and younger and 17.7% of the time in women over 35, suggesting that menstruation is also affected by shifts in the moon’s gravimetric forces. Furthermore, the researchers observed greater synchronization between lunar and menstrual cycles during long winter nights, when women experienced prolonged exposure to moonlight. While the moon’s luminescence and gravimetric cycles each appeared to only weakly affect menstrual cycles on their own, the findings suggest that these cycles exhibit a stronger effect together, with menstrual cycles most in sync with lunar rhythms when the moon is nearest to the Earth.
